Pemetrexed is a chemotherapy medicine used mainly to treat non-small cell lung cancer and malignant pleural mesothelioma. It belongs to a group of medicines called antimetabolites, which interfere with the growth and spread of cancer cells by blocking the enzymes they need to make DNA and RNA.
Pemetrexed works by inhibiting several key enzymes involved in cell replication, including thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyltransferase. Less commonly, pemetrexed may cause lung inflammation, which can present as shortness of breath or cough. Any new or worsening breathing symptoms should be reported immediately.
Pemetrexed can harm an unborn baby and must not be used during pregnancy. Effective contraception is required during treatment and for a period afterward. Breastfeeding is not recommended during treatment.
Patients receiving pemetrexed should avoid non-steroidal anti-inflammatory drugs (such as ibuprofen) around the time of treatment unless approved by their healthcare provider, as these can affect drug clearance and increase side effects.
